Submission Requirements:

Our whole plasmid sequencing service requires you submit samples that meet quality and quantification requirements. The best results will come from whole, double stranded, clonal plasmids of high purity and an accurate concentration. Standard plasmid prep methods that have been performed correctly should be able to meet these requirements.

Purified plasmids should be in 10mM Tris of nuclease-free water.

A nanodrop reading with a 260/280 of <1.8 and a 260/230 of <2.0-2.2 indicates the sample is free from contaminants that will inhibit a successful sequencing run.

Visualizing the plasmid DNA on an agarose gel can confirm there is a single plasmid present at the expected size.

A fluorometric reader like the Qubit or equivalent give the most accurate reading of concentrations – a concentration based on other methods, such as spectrophotometers, may lead to a miss-estimation of DNA quantity.
